The Science And Technology Of Human Pheromones

The chemistry:

The bulk majority of human pheromones belong to a group of chemicals known as 16-androstenes. The majority of active human pheromones have an unsaturated double bond at the 16th carbon of the molecular structure. However, there are active pheromones which lack this group, such as our proprietary androstanone, and are known as androstanes. Androstanes are more polar than androstanes, and hence are hygroscopic and water soluble (this is inherently interrelated to increased efficacy at receptor sites via increased membrane permeability).

All known human pheromones contain an oxygen atom bound to the 3rd carbon in A-ring: such as in the form of a keto, or an alcohol, acetate, sulfate, or other functional groups. The keto or hydroxy groups are most commonly found at this position. Compounds with 3-OH groups tend to have stronger effects upon regulation of currents within the hypothalamus.

Compounds which have ionic functional groups at this position have superior performance versus those without. It appears as though this is due to the greater water-solubility and hence targeting affinity for the receptor sites within the olfactory bulb and accessory olfactory system. The enhanced electronegativity of the molecule may also enhance the affinity for the chemosensory receptors.

There are several compounds known which have functional groups at the 17th carbon in D-ring. Depending on the type of functional group, this substitution may either decrease or increase the performance of the pheromone significantly.17-one or 17-ol groups tend to reduce performance, whereas 17-Carboxyl groups may enhance performance, and whereas allene or aromatic groups may have great increases in performance, with a significant change in the character of the parent compound.

Pheromones fit into a category of chemical sensing that is very similar to the way we smell, called olfaction. However, it is believed that pheromones are detected and processed by an accessory olfactory system. In mammals, this is the Vomeronasal System. For over 200 years, scientific and medical researchers had suspected that airborne chemical compounds played some role in the attraction and mating rituals of insects and animals. A Danish physician named Jacobson even detected likely organs for this purpose in the human nose, but he assumed that evolution had rendered them non-functional.

It took German researchers extracting glands from the abdomens of 500,000 female moths over a 20-year period to isolate this compound, called bombykol. "It has been soberly calculated that if a single female moth were to release all the bombykol in her sac in a single spray, all at once, she could theoretically attract a trillion males in the instant," wrote Lewis Thomas in The Lives of a Cell.

Isolating pheromones in mammals (which includes you and me) proved to be a much more complex challenge.

But by the late 1970s, animal pheromones had become well enough understood to market commercially as a lure to attract and divert animals into traps before they could damage crops or flowers. Finally, in 1986, sufficient research had been done on the subject of human pheromones for peer-reviewed articles to be published in respected scientific journals.
